Detecting marker-disease association by testing for Hardy-Weinberg disequilibrium at a marker locus.
We review and extend a recent suggestion that fine-scale localization of a disease-susceptibility locus for a complex disease be done on the basis of deviations from Hardy-Weinberg equilibrium among affected individuals. This deviation is driven by linkage disequilibrium between disease and marker loci in the whole population and requires a heterogeneous genetic basis for the disease. A finding...
متن کاملSurvival quantitative trait locus fine mapping by measuring and testing for Hardy-Weinberg and linkage disequilibrium.
I show that fine-scale localization of a survival-related locus can be accomplished on the basis of deviations from Hardy-Weinberg equilibrium and linkage disequilibrium at closely linked marker loci. The method is based on chi(2)-tests and they can be performed for age-specific samples of alive (or dead) individuals, as for combined samples of alive and dead individuals.
متن کاملLosing balance: Hardy-Weinberg disequilibrium as a marker for recurrent loss-of-heterozygosity in cancer.
Identifying regions of loss-of-heterozygosity (LOH) in a tumor sample is a challenging problem. State-of-the-art computational approaches can infer LOH from single-nucleotide polymorphism (SNP) array data, but calling precise boundaries is complicated by normal-cell contamination and markers that are homozygous in the germline and therefore non-informative. More recently, the focus has shifted ...
متن کاملLee Responds to “Testing for Hardy-Weinberg Disequilibrium”
I appreciate Weinberg and Morris’ thoughtful commentary (1) on my paper (2). In their article, they put my work under the perspective of gene mapping in the postgenomic era. I share the same view with them that the method proposed in my paper amounts to a tree-shaking approach to harvesting the high-hanging fruit (a low-cost approach to generating hypotheses aimed at localizing diseasesusceptib...
متن کاملQTL fine mapping by measuring and testing for Hardy-Weinberg and linkage disequilibrium at a series of linked marker loci in extreme samples of populations.
It has recently been demonstrated that fine-scale mapping of a susceptibility locus for a complex disease can be accomplished on the basis of deviations from Hardy-Weinberg (HW) equilibrium at closely linked marker loci among affected individuals. We extend this theory to fine-scale localization of a quantitative-trait locus (QTL) from extreme individuals in populations, by means of HW and link...
